Oxidation of cadaverine by putrescine oxidase from Rhodococcus erythropolis

چکیده
BACKGROUND Putrescine oxidase (EC 1.4.3.10) is of interest for the microbial production unsubstituted platform nitrogen (N-)heterocycles, because it only requires inexpensive oxygen as co-substrate. from Rhodococcus erythropolis (Re-PuO) was shown previously to catalyze oxidation cadaverine; however, there little information in literature about robustness this enzyme biotechnological applications. The aim study investigate suitability Re-PuO bioproduction 1-piperideine cadaverine under different reaction conditions. RESULTS formation catalyzed by demonstrated using o-aminobenzaldehyde a reagent trap cyclic imine and shift equilibrium cyclization. A direct assay activity then implemented, monitoring consumption. Characterization mixture 1H NMR mass spectrometry confirmed presence piperideine dimers trimers, yet quantification products could not be achieved. optimum temperature pH conditions were determined 55 °C 8.5, respectively. At 7.5, retained its after 65 h incubation at 25 °C, but lost 75% 1 °C. showed no substrate inhibition concentrations high 100 mmol L–1 cadaverine. Complete biotransformation observed whole cells physiological CONCLUSIONS These results successfully demonstrate potential putrescine N-heterocycles © 2021 Authors. The complete alk sequences of Rhodococcus erythropolis from Lake Baikal
BACKGROUND Rhodococci are bacteria able to degrade a wide range of hydrocarbons, including the alkanes present in crude oil, due to alk genes in their genomes. FINDINGS Genome sequencing of DNA from Rhodococcus erythropolis strain 4 (obtained from a deep-water bitumen mound) revealed four alk genes, and the predicted amino acid sequences coded by these genes were highly conserved, having sect...
متن کاملCharacterization of catechol catabolic genes from Rhodococcus erythropolis 1CP.
The biochemical characterization of the muconate and the chloromuconate cycloisomerases of the chlorophenol-utilizing Rhodococcus erythropolis strain 1CP previously indicated that efficient chloromuconate conversion among the gram-positive bacteria might have evolved independently of that among gram-negative bacteria. Based on sequences of the N terminus and of tryptic peptides of the muconate ...
متن کاملExtracellular Cholesterol Oxidase from Rhodococcus sp.: Isolation and Molecular Characterization
Background: Cholesterol oxidase (CHO) has various clinical and industrial applications. Recently, microbial CHO have received a great attention for their wide usage in medicine. Here, taxonomic characterizations of isolated strain from soil, optimization of the conditions for CHO production and biochemical characterizations of produced CHO enzyme were described. Finally, CHO gene was cloned int...
متن کاملRhodococcus erythropolis Encephalitis in Patient Receiving Rituximab
To the Editor: Rhodococcus spp. infections occur predominantly in immunocompromised patients, and most infections are caused by Rhodococcus equi (1). Seven cases of R. erythropolis infections in humans have been described (2–8). None of these cases included a central nervous system infection (Table). We report a case of R. erythropolis meningoencephalitis in a patient with rheumatoid arthritis ...
متن کاملProduction of Biosurfactant by Hydrocarbon Degrading Rhodococcus Ruber and Rhodococcus Erythropolis
There is world wide concern about the liberation of hydrocarbons in the environment, both from industrial activities and from accidental spills of oil and oil-related compounds. Biosurfactants, which are natural emulsifiers of hydrocarbons, are produced by some bacteria, fungi and yeast. They are polymers, totally or partially extracellular, with an amphipathyc structure, which allows them to f...
